May I know how can I update drop down option in spreadsheet - > appsheet detect changes and sync without pressing regeneration in AppSheet browser?

The only option to detect changes to data validation rules in the spreadsheet is to regenerate columns in the app editor.

Instead of using spreadsheet data validation, is there any in app formua that can update itself and have the same function as data validation?

It depends on how you’re using data validation. If you’re just using it to present and enforce a list of choices, you can use an Enum or EnumList column within AppSheet.
